What bacteriostatic water is, why laboratories use it to reconstitute lyophilised peptides, and how to store it.
Mechanism of Action
Bacteriostatic water is not a peptide and has no pharmacological mechanism of its own. It is a diluent: sterile water containing approximately 0.9% benzyl alcohol as a preservative.
Benzyl alcohol is bacteriostatic rather than bactericidal — it inhibits the multiplication of bacteria that may be introduced into the vial rather than sterilising the contents outright. That distinction matters in practice: the preservative extends the usable life of a multi-draw vial, but it does not substitute for aseptic technique.
Research Applications
In laboratory settings, bacteriostatic water is the standard reconstitution solvent for lyophilised research peptides. Lyophilised material is stable as a dry cake but must be brought into solution before it can be measured, diluted or applied in an assay, and a preserved diluent allows a single vial to be drawn from more than once over a working period without the microbial risk of unpreserved sterile water.
It is also used for preparing intermediate dilutions and for rinsing and equilibrating glassware in workflows where the preservative is not an interfering variable. Where benzyl alcohol could interfere with an assay — certain cell-viability and microbiological readouts in particular — unpreserved sterile water or an appropriate buffer should be used instead, and the choice recorded in the method.
Storage & Stability (Laboratory Information)
Sealed vials are stable at controlled room temperature, away from direct light and heat. Freezing is unnecessary for the diluent and is not recommended.
Vial contents should be clear and colourless with an intact seal and no visible particulates; access and handling are governed by the receiving institution's own aseptic technique and COSHH procedures.
An entered vial is a multi-use container with a limited working life. Laboratory practice commonly references up to 28 days from first entry when stored correctly, though the batch documentation supplied with the vial takes precedence. Any vial that becomes cloudy, discoloured or particulate, or whose stated in-use period has elapsed, is treated as waste.
