Tesamorelin: A Stabilised GHRH Analog in Endocrine Research

A modified growth-hormone-releasing hormone analog used to study pituitary GHRH receptor signalling in research models.

Mechanism of Action

Tesamorelin is a stabilised analog of growth-hormone-releasing hormone (GHRH). Native GHRH is rapidly degraded by dipeptidyl peptidase enzymes; the analog carries a structural modification at the N-terminus that has been reported to slow that degradation, giving a longer-lived research tool for studying the same receptor. All activity described below is reported from in vitro and animal-model work.

The mechanism examined is engagement of the pituitary GHRH receptor, a G-protein-coupled receptor. In cell-based models the endpoints measured are intracellular cyclic AMP accumulation and growth hormone content in the medium of somatotroph preparations. Because the compound acts on the receptor rather than substituting for growth hormone, it is used as a probe of receptor pharmacology and of feedback within that axis in model systems.

In model systems, typical readouts include cAMP accumulation in pituitary cell preparations, growth hormone secretion profiles over time, and the pulsatility of that secretion compared with native GHRH.

Research Applications

Tesamorelin is used in endocrine research examining GHRH receptor pharmacology, somatotroph signalling and the regulation of the growth hormone axis.

It also appears in endocrine model systems where growth hormone axis markers are measured alongside lipid-handling assays in adipose tissue preparations; the recorded variables are analyte concentrations, not outcomes.

Storage & Stability (Laboratory Information)

Tesamorelin is supplied as a lyophilised powder in a sealed amber vial. Lyophilised peptide material is generally most stable when the sealed vial is kept frozen and protected from light; condensation on cold glass is a known source of moisture ingress, which is why sealed vials are commonly equilibrated to ambient temperature before a seal is broken.
